20080280779 | Gene expression profiling based identification of genomic signatures of multiple myeloma and uses thereof - Monoclonal gammopathy of undetermined significance can progress to multiple myeloma. Applying significance analysis of microarrays, 52 genes, involved in important pathways related to cancer, were differentially expressed between plasma cells from healthy subjects and patients with stringently defined monoclonal gammopathy of undetermined significance/smoldering multiple myeloma and symptomatic multiple myeloma. Unsupervised hierarchical clustering of 351 multiple myeloma and 44 cases of monoclonal gammopathy of undetermined significance and 16 cases of multiple myeloma with a monoclonal gammopathy of undetermined significance history, created two major cluster branches, one containing 82% of the monoclonal gammopathy of undetermined significance cases and 28% of the multiple myeloma, termed monoclonal gammopathy of undetermined significance-like multiple myeloma. Using the same clustering approach on an independent cohort of 213 cases of multiple myeloma revealed 27% with monoclonal gammopathy of undetermined significance-like multiple myeloma which, despite a lower incidence of complete remission, was associated with low-risk clinical and molecular features and superior survival. The monoclonal gammopathy of undetermined significance-like multiple myeloma signature was also seen in patients surviving more than 10 years after autotransplant. | 11-13-2008 |
20100144673 | Diagnosis, prognosis and identification of potential therapeutic targets of multiple myeloma based on gene expression profiling - Gene expression profiling is a powerful tool that has varied utility. It enables classification of multiple myeloma into subtypes and identifying genes directly involved in disease pathogensis and clinical manifestation. The present invention used gene expression profiling in large uniformly treated population of patients with myeloma to identify genes associated with poor prognosis. It also demonstrated that over-expression of CKS1B gene, mainly due to gene amplification that was determined by Fluorescent in-situ hybridization to impart a poor prognosis in multiple myleoma. It is further contemplated that therapeutic strategies that directly target CKS1B or related pathways may represent novel, and more specific means of treating high risk myeloma and may prevent its secondary evolution. | 06-10-2010 |
20100152136 | TP53 Gene expression and uses thereof - The present invention is drawn to diagnosis, prognosis and treatment of multiple myeloma. In this regard, the present invention discloses importance of down-regulation of TP3 gene in multiple myeloma and its use as an independent progostic indicator of multiple myeloma. Additionally, the present invention also discloses novel-TP53 associated genes and demonstrates the clinical relevance of these alterations to disease progression. | 06-17-2010 |
20100316629 | Use of gene expression profiling to predict survival in cancer patient - Gene expression profiling in multiple myeloma patients identifies genes that distinguish between patients with subsequent early death or long survival after treatment. Poor survival is linked to over-expression of genes such as ASPM, OPN3 and CKS1B which are located in chromosome 1q. Given the frequent amplification of 1q in many cancers, it is possible that these genes can be used as powerful prognostic markers and therapeutic targets for multiple myeloma and other cancer. | 12-16-2010 |
20110269638 | Genes associated with post relapse survival and uses thereof - Provided are methods, systems and kits for predicting post-relapse survival of a cancer patient and for identifying cancer genes predictive of the post-relapse survival of the patient. Values representing gene expression levels of a group of genes associated with survival of the cancer cells are determined using gene expression profiling platforms and a plurality of probe sets that hybridize to one or more of the genes in the group. A predictive model establishes a predictive value based on the weighted contribution of each gene associated with survival of the cancer cells to risk of death for the cancer patient and imports expression values of the genes in the group that is indicative of a risk of death for the relapsed patient. Using global gene expression profiling and statistical analysis, expression of cancer cell genes at baseline and at first relapse that are involved in interaction of cancer cells with cells in their microenvironment, can be used to identify genes that are predictive of post-relapse survival. | 11-03-2011 |
20120015906 | USES OF BORTEZOMIB IN PREDICTING SURVIVAL IN MULTIPLE MYELOMA PATIENTS - The present invention provides a method of predicting outcome of treatment for multiple myeloma based on determining certain cytogenetic anomalies and considering gene expression profiling risks. Also provided are statistical methods employed to define variables independently impacting outcomes. Further provided is method of treatment of myeloma patients. | 01-19-2012 |
20120269802 | Treatment And Prognosis With Thalidomide In Multiple Myeloma Based on Karyotyping And Gene Expression Profiling - The present invention provides a method treating a myeloma patient by administering one or more of thalidomide, a Total Therapy 2 regimen, an interleukin-6 signaling suppressor, an interleukin-6R signaling suppressor, an IGF1 signaling suppressor, an IGF1 R signaling suppressor, shRNA or other modulators of gene expression. Also, provided are methods for predicting outcome of a treatment for an individual having a cancer, e.g., myeloma, by performing one or more of karyotyping or expression profiling of chromosomes 1 and 13 or expression level measurement of IL-6R. | 10-25-2012 |
20130059746 | GENE EXPRESSION PROFILING OF CYTOGENETIC ABNORMALITIES - Provided herein are methods of predicting cytogenetic abnormalities associated with a cancer in a subject, for example, multiple myeloma. A cytogenetic abnormalities model of a set of reference values obtained from an average of gene expression profile values based on copy number-sensitive genes that correlate to cytogenetic abnormalities associated with the cancer is utilized as a predictive tool. The cytogenetic abnormalities model, as a virtual model (i.e. a “virtual karyotype”), may be tangibly stored with program instructions to implement the model in a computer system. In particular embodiments, the methods and systems provided by the invention operate without FISH (fluorescent in situ hybridization). | 03-07-2013 |
20130072392 | Compositions, Kits, and Methods for Identification, Assessment, Prevention, and Therapy of Cancer - The invention relates to compositions, kits, and methods for detecting, characterizing, preventing, and treating human cancer. A variety of chromosomal regions (MCRs) and markers corresponding thereto, are provided, wherein alterations in the copy number of one or more of the MCRs and/or alterations in the amount, structure, and/or activity of one or more of the markers is correlated with the presence of cancer. | 03-21-2013 |
20130209446 | Copy Number Variant-Dependent Genes As Diagnostic Tools, Predictive Biomarkers And Therapeutic Targets - Provided herein are methods of diagnosing and/or treating malignant or pre-malignant conditions in a subject. Overexpression of copy number variant-dependent genes, e.g., genes encoding a cell surface receptor, resulting from copy number changes compared to control is diagnostic of the condition, such as multiple myeloma or monoclonal gammopathy of undetermined significance. Also provided are methods for treating malignant conditions, such as multiple myeloma or a hyperdiploid subtype, with therapeutic agents, with or without other anti-cancer drugs, to downrregulate the overexpressed CNV genes and/or up-regulate the underexpressed genes. Furthermore, methods for lowering drug resistance in multiple myeloma cells via inhibition of platelet activation or thrombin release and for increasing survivability of a multiple myeloma subject via lithibition of PSMD4 gene to increase b-catenin protein expression are provided. | 08-15-2013 |
